Organization Processing

1.Take fresh tissue thickness not more than 5mm, 10% neutral formalin fixation, more than 24 hours.

2.After fixation, flush for 12-24 hours.

3.75% alcohol, once, one hour.

4.85% alcohol, once, one hour.

5.95% alcohol, 3 times, one hour.

6.100% alcohol, 3 times, 1 hour.

7.Xylene, 2 times, 1 hour.

8.Paraffin soak, 3 times, 2 hours.

HE Dyeing

(Hydration)

1.Xylene, 2 times, 5-10 minutes.

2.100% alcohol, once, 1-2 minutes.

3.95% alcohol, once, 1-2 minutes.

4.85% alcohol, once, 1-2 minutes.

5.75% alcohol, once, 1-2 minutes.

6.Over distilled water.

(Dyeing)
1.Mayer's hematoxylin, 1 minute.

2.Rinse with warm water， 5-10 minutes.

3.75% hydrochloric alcohol, 1-2 minutes.

4.Tap water rinse, 30 seconds.

5.Reddening, 1-2 minutes.

6.Tap water, 30 seconds.

7.85% alcohol, once, 1-2 minutes.

8.95% alcohol, 2 times, 1-2 minutes.

9.100% alcohol, 2 times, 1-2 minutes.

10.Xylene, 2 times, 5-10 minutes.

11.Neutral gum sealing piece.
